Ivacaftor is a potentiator of the CFTR protein.

The CFTR protein is a chloride channel present at the surface of epithelial cells in multiple organs.

Ivacaftor is extensively metabolized in humans.

How it leaves the body

Following oral administration, the majority of ivacaftor (87.8%) is eliminated in the feces after metabolic conversion.

With food

The exposure of ivacaftor increased approximately 2.5- to 4-fold when given with food that contains fat.

Most common side effects

The most common adverse reactions in the 221 patients treated with KALYDECO were headache (17%), upper respiratory tract infection (16%), nasal congestion (16%), nausea (10%), rash (10%), rhinitis (6%), dizziness (5%), arthralgia (5%), and bacteria in sputum (5%).
