How it works
From the US prescribing label for the same active substance (Tacrolimus). It describes the substance, not this exact product.
Tacrolimus binds to an intracellular protein, FKBP-12.
A complex of tacrolimus-FKBP-12, calcium, calmodulin, and calcineurin (a ubiquitous mammalian intracellular enzyme) is then formed, after which the phosphatase activity of calcineurin is inhibited.
Tacrolimus is extensively metabolized by the mixed-function oxidase system, primarily the cytochrome P-450 system (CYP3A4 and CYP3A5).
In man, less than 1% of the dose administered is excreted unchanged in urine.
Food Effects The rate and extent of tacrolimus absorption were greatest under fasted conditions.
